بررسی تأثیر افزودن ویتامین ای (آلفا توکوفرول) به رقیق‌کننده بر کیفیت اسپرم منجمد سگ

چکیده
انجماد اسپرم فرایندی پیچیده است که اشکال مختلفی از آسیب‌های سلولی را با خود به همراه دارد. چنین آسیب‌هایی ممکن است در اسپرم ذوب شده مورد استفاده برای تلقیح مصنوعی باعث کاهش تحرک و باروری شود. در این پژوهش پانزده نمونه منی از پنج قلاده سگ نر جمع‌آوری و هر نمونه جداگانه به سه قسمت (کنترل، دارای 2/0 میلی‌مولار ویتامین E و دارای 4/0 میلی‌مولار ویتامین E) تقسیم شدند. نتایج این مطالعه نشان داد که افزایش معنی‌دار در تحرک اسپرم‌های روبه‌جلو با سرعت بالا اتفاق افتاده است (05/0≤ P). در این پژوهش افزایش معنی‌دار در تحرک اسپرم‌های روبه‌جلو با سرعت بالا و همچنین مقادیر VCL، VSL، VAP و BCF در گروه‌های 2/0 و 4/0 نسبت به گروه کنترل، تنها در گروه مقاوم در برابر شوک سرمایی رخ‌داده است (05/0≤ P). در آزمون ارزیابی یکپارچگی غشای اسپرم در گروه 4/0 نسبت به گروه کنترل افزایش معنی‌دار وجود داشت (05/0≤ P). این مطالعه نشان داد که استفاده از ویتامین E با غلظت‌های 2/0 و 4/0 میلی‌مولار در ترکیب رقیق‌کننده اسپرم منجمد سگ مفید واقع می‌شود. همچنین در این پژوهش مشخص شد تنها اسپرم‌های مقاوم در برابر شوک سرمایی (نسبت به اسپرم‌های حساس به این پدیده) از اثرات مثبت ویتامین E برخوردار می‌شوند و می‌توان فرض کرد که این موضوع به دلیل مقادیر بالاتری از اسید های چرب غیراشباع در معرض پراکسیداسیون در این گروه از اسپرم‌ها باشد.

Evaluation of the Effects of Vitamin E (α-Tocopherol) Supplementation in the Extender Containing Egg Yolk on the Characteristics of Canine Cryopreserved Semen

Freezing sperm is a complex process that can result in various forms of cell damage. Such damage may reduce the motility and fertility of thawed sperm used for artificial insemination. In this study, fifteen semen samples were collected from five mixed-breed male dogs, and each sample was divided into three parts (control, 0.2 mmol vitamin E, and 0.4 mmol vitamin E). Although motility was observed in groups that received antioxidants, this increase was not statistically significant. However, the average motility of high-speed progressive sperm after thawing indicates a significant increase (P≤0.05) in their motility. The study showed a significant increase (P≤0.05) in high-speed progressive sperm motility, VCL, VSL, VAP, and BCF values in groups 0.2 and 0.4 compared to the control group. However, this increase was only observed in the cold shock-resistant group. In the test, the evaluation of the integrity of the sperm membrane in Group 0.4 showed a significant increase (P≤0.05) compared to the control group. The study finds that using vitamin E with concentrations of 0.2 and 0.4 improves the resistance of sperm to shocks compared to sperm that are sensitive to this phenomenon. This positive effect of vitamin E can be attributed to higher levels of unsaturated fatty acids that are susceptible to peroxidation in this group of sperm.
