مقایسه شاخص‌های قابل اندازه‌گیری توسط کاسا در نمونه‌های اسپرم گاوهای نر سیمنتال همراه‌شده با غلظت‌های متفاوت نانوکورکومین

چکیده
زردچوبه (Curcuma longa) گیاهی چندساله متعلق به خانواده زنجبیل است. فراهمی زیستی ضعیف کورکومین به‌دلیل جذب پایین، سوخت و ساز سریع و حذف سیستمیک سریع از بدن و هم‌چنین میزان حلالیت کم، مشکلی مهم در استفاده از آن است. یکی از پیشنهادات به‌منظور بهبود فراهمی زیستی کورکومین، استفاده از نانوذرات آن است. مشخص شده که کورکومین اثرات آنتی‌اکسیدانی چشم‌گیری داشته و استفاده از آن سبب بهبود فعالیت تولیدمثلی جنس نر می‌شود. بنابراین، هدف این مطالعه بررسی اثرات اضافه‌کردن نانوکورکومین به رقیق‌کننده اسپرم بر پارامترهای کیفی منی در اسپرم‌های تازه و یخ‌گشایی شده در گاوهای نر سیمنتال دومنظوره (Fleckvieh) بود. نمونه‌های منی از 12 گاو نر در سه تکرار در بازه زمانی سه‌ماهه اخذ شد. هر انزال به‌طور مساوی به سه قسمت برای گروه‌های آزمایشی (کنترل، نانوکورکومین 50 میلی‌گرم در لیتر و نانوکورکومین 100 میلی‌گرم در لیتر) تقسیم شد. انجماد اسپرم به‌روش رقیق‌سازی دومرحله‌ای انجام شد. پارامترهای کیفی اسپرم شامل تحرک، زنده‌مانی، شکل‌شناسی و یک‌پارچگی غشا قبل و بعد انجماد بررسی شدند. نتایج یافته‌ها نشان داد که نانوکورکومین بر کیفیت اسپرم‌های قبل از انجماد تأثیر مثبتی نداشت، اما سبب افزایش تحرک و پارامترهای تحرک در گروه 100 میلی‌گرم در لیتر در اسپرم‌های منجمد یخ‌گشایی شده گردید. از آنجایی‌که تحرک پیش‌رونده پس از انجماد از مهم‌ترین شاخص‌های تخمین کیفیت اسپرم‌ است، پیشنهاد می‌شود نانوکورکومین به میزان 100 میلی‌گرم به رقیق‌کننده اسپرم گاوهای نر اضافه شود و پژوهش‌های برون‌تنی و درون‌تنی جهت تعیین میزان باروری و نرخ آبستنی در تخمک‌های بارورشده و گاوهای ماده لقاح‌یافته با اسپرم‌های غنی‌شده با نانوکورکومین، انجام شود.

چکیده English

Turmeric (Curcuma longa) is a perennial plant belonging to the ginger family. Poor bioavailability of curcumin due to low absorption, fast metabolism and systemic removal as well as low solubility are important problems in optimal use of curcumin. One of the suggestions to improve the bioavailability of curcumin is to use its nanoparticles. It has been found that curcumin has significant antioxidant effects and can improve male reproductive activity. Therefore, the aim of this study was to investigate the effects of adding nanocurcumin to semen extender on the semen quality in fresh and frozen-thawed spermatozoa in dual purpose Simmental bulls (Fleckvieh). Ejaculates were collected from 12 bulls in three repetitions in a period of three months. Each ejaculate was equally divided into three parts for experimental groups (control, nanocurcumin 50 mg/L and nanocurcumin 100 mg/L). Semen was frozen in a two-step dilution method. Sperm parameters including motility, viability, morphology, and membrane integrity were investigated before and after freezing. The results showed that nanocurcumin did not have a positive effect on the quality of fresh sperms while, caused an increase in sperm motility parameters in the 100 mg/L group in thawed semen. Since sperm progressive motility of thawed semen is one of the most important indicators of sperm quality prediction, it is suggested to add 100 mg/L of nanocurcumin to the bull semen extenders and conduct in vitro and in vivo studies to determine the fertility and conception rate in fertilized oocytes and cows inseminated with thawed semen enriched with nanocurcumin.
