مقایسه فصلی هیستومورفولوژی و تغییرات میزان رونویسی ژن IGF-1 روده گاوهای منطقه شهرکرد

چکیده
فاکتور رشد شبه انسولین 1 (IGF-1) همراه هورمون رشد (GH) اثرات تحریکی بر تکثیر غضروف و استخوان دارند، اما اطلاعات کمی در مورد نقش IGF-1 در رشد پرزهای روده کوچک طی فصول سرد و گرم سال وجود دارد. بر این اساس مطالعه حاضر جهت تعیین تغییرات فصلی میزان نسخه‏برداری از ژن IGF-1، تفاوت رشد فصلی کریپت‏ها و پرزهای روده کوچک و سطوح سرمی روی و منیزیم انجام شد. بدین منظور از 30 رأس گاو ذبح شده بیشتر از 3 سال سن، طی فصول زمستان و تابستان نمونه خون و بافت روده (دوازدهه) گرفته شد. غلظت روی و منیزیم سرم با استفاده از کیت تجاری و برای تعیین میزان نسخه‏برداری از ژن IGF-1 روده از روش RT-PCR استفاده شد. اندازه‌گیری‌های هیستومورفولوژیک نیز با استفاده از نرم‌افزار Image tools III مورد ارزیابی قرار گرفت. بر اساس نتایج به‌دست‌آمده، نرخ رونویسی ژن IGF-I و اندازه پرزها در فصل زمستان به ترتیب 82/1 و 2/1 برابر تابستان بود (05/0P<). برعکس، غلظت منیزیم در تابستان به طور قابل توجهی بیشتر از زمستان بود (05/0P<). در حالی که از نظر آماری، تفاوت معنی داری در سطح سرمی روی مشاهده نشد (05/0P>). یافته‏ها نشان دهنده وجود همبستگی مثبت میان میزان رونویسی ژن IGF-1 و نرخ رشد پرزهای روده بود (0341/0 P= و 852/0+ r=). در نتیجه، تغییر فصلی در میزان رونویسی ژن IGF-1 برای اولین بار در روده کوچک گزارش شد. همچنین، وجود هیپومنیزیمی در زمستان را می‏توان به عنوان یک عامل خطر برای مستعد ساختن گاو به هیپوکلسمی تحت بالینی و بالینی در نظر گرفت.

Insulin-like growth factor 1 (IGF-1) and growth hormone (GH), exerts stimulatory effects on cartilage and bone proliferation; however, limited information exists regarding the role of IGF-1 in small intestinal villus growth during cold and warm seasons. Accordingly, the present study was conducted on slaughtered dairy cows to determine the seasonal variations of serum zinc and magnesium levels, IGF-1 gene transcription rate and seasonal variations in small intestine crypt and villus size. To these reasons, blood and intestinal (duodenal) tissue samples were collected from 30 slaughtered cattle (aged over three years) during the winter and summer seasons. Serum zinc and magnesium concentrations were measured using commercial kits, and RT-PCR was employed to determine the intestinal IGF-1 gene transcription rate. Histomorphometric measurements were also evaluated using Image Tools III software. The results indicated that the IGF-1 gene transcription rate and villus size in winter were 1.82 and 1.2 times those observed in summer, respectively. Conversely, magnesium concentration was significantly higher in summer than in winter (P<0.05), whereas no statistically significant difference was observed in serum zinc levels. These findings indicated a positive correlation between intestinal IGF-1 gene transcription rate and the growth rate of intestinal villi. Consequently, seasonal variation in intestinal IGF-1 gene transcription rate has been reported here for the first time. Furthermore, the occurrence of hypomagnesemia in winter should be considered a risk factor predisposing cattle to subclinical and clinical hypocalcemia.
